Amine reactive Cyanine7, near infrared fluorescent dye, an improved analog of Cy7®.

NIR fluorophores can be used to take advantage of near infrared window of biological tissues – increased transparency of tissues in this spectral region allows to carry out in vivo imaging.

This reagent can be utilized to produce Cyanine7-labeled biomolecules for subsequent use in various in vivo research, and drug design related experiments.

The structure of Cyanine7 features rigidized design of central polymethyne chain. This molecular reinforcement allows to increase quantum yield by 20% compared with parent structure, increasing fluorescence brightness.

This reagent requires organic co-solvent for the labeling (please see Recommended Protocols section below). Water-soluble Cyanine7 NHS ester is also available, and recommended for protein NIR labeling.

There are two varieties of cyanine dyes: non-sulfonated cyanines, and sulfonated cyanines. For many applications they are interchangeable, because their spectral properties are nearly identical. Both sulfonated and non-sulfonated dyes can be used for the labeling of biomolecules such as DNA and proteins. The difference between the dyes is their solubility: sulfo- dyes are water-soluble, and they do not use of organic co-solvent for the labeling in aqueous environment. They are less prone to aggregation in water.

The reagent is useful as a fluorescent marker in NIR range when attachment to other molecules is not desired. It has high hydrophilicity and aqueous solubility, improved quantum yield in NIR range, and very high molar extinction coefficient.

This reagent contains free carboxylic acid function. Pre-activated activated ester – sulfo-Cyanine7 NHS ester is also available.

This is a reagent of choice for the labeling of antibodies, sensitive proteins, and others which require reactions in purely aqueous environment, or without any significant additions of organic co-solvents.

The dye is well suitable for non-invasive in vivo NIR imaging, and for the applications requiring low fluorescent background.
